RESUMO

OBJECTIVE To determine the contents of 11 components in Xueli zhike syrup, establish its chemometric method and provide reference for its quality control. METHODS HPLC method was established to simultaneously determine the contents of amygdalin, deapi-platycoside E, platycoside E, platycodin D3, euscaphic acid, tormentic acid, maslinic acid, corosolic acid, praeruptorin A, praeruptorin B and praeruptorin E in 12 batches of Xueli zhike syrup. The quality evaluation of 12 batches of samples was performed by chemometrics. RESULTS The 11 components had good linear relationships within their respective ranges (r≥0.999 1); RSDs of precision, reproducibility and stability (24 h) tests were all lower than 2.00%. The average recovery rates ranged 96.90%-100.01% (RSDs were all lower than 2.00%). Cluster analysis showed that 12 batches of samples were clustered into 3 groups. Principal component analysis showed that the first two principal components could represent 88.53% information of 11 components in Xueli zhike syrup. Partial least squares-discrimination analysis showed that euscaphic acid, amygdalin and praeruptorin A were the main potential markers affecting the quality of Xueli zhike syrup. CONCLUSIONS The established method can be used to control the quality of Xueli zhike syrup.

RESUMO

Objective:To systematically evaluate the relationship between CXCR4 and the clinicopathological characteristics and prognosis of patients with non-small cell lung cancer (NSCLC).Methods:The PubMed, EMbase, Cochrane Library, CNKI, Wanfang and China Biology Medicine databases were searched by the computer to collect the publicly published cohort studies on the correlation between CXCR4 expression and prognosis of NSCLC. The retrieval time was from the establishment of the database to May 31, 2018. Literature screening, data extraction and risk of bias were performed independently by two reviewers, and Meta-analysis was performed by using Stata 12.0 software. Results:Finally, 20 studies were included, with a total of 2 124 NSCLC patients. The positive expression rate of CXCR4 in patients with adenocarcinoma was 51.8% (337/650) and it was 50.3% (224/445) in patients with squamous cell carcinoma, the difference was statistically significant ( RR=1.22, 95% CI 1.09-1.38, P=0.001). The positive expression rate of CXCR4 in patients with stageⅠ-Ⅱ was 41.1% (317/772), and the rate was 57.0% (348/610) in patients with stage Ⅲ-Ⅳ, the difference was statistically significant ( RR=0.62, 95% CI 0.55-0.69, P < 0.01). The positive expression rate of CXCR4 in patients with distant metastasis was 55.3% (268/485), and the positive rate of CXCR4 in those without distant metastasis was 39.2% (256/653), the difference was statistically significant ( RR=1.78, 95% CI 1.54-2.06, P < 0.01). The expression of CXCR4 was not related to the size and invasion of the primary tumor, the positive expression rates of CXCR4 in T 1-T 2 and T 3-T 4 patients were 41.0%(215/524) and 53.8%(84/156) ( RR=0.71, 95% CI 0.60-0.84, P=0.053). The overall survival rate of patients with CXCR4-positive was lower than that of patients with CXCR4-negative, and the difference was statistically significant (30.1% vs. 66.9%, RR=0.45, 95% CI 0.30-0.61, P < 0.01). The disease-free survival rate of patients with CXCR4-positive was lower than that of patients with CXCR4-negative (38.0% vs. 20.1%, RR=1.89, 95% CI 1.45-2.47, P=0.002). Conclusions:The pathological type, tumor staging and distant metastasis of NSCLC patients are correlated with CXCR4 expression level. The expression level of CXCR4 in tumor tissues has a predictive value for the prognosis of NSCLC patients; and the higher the expression level of CXCR4 is, the lower the overall survival and disease-free survival rates are.

RESUMO

Objective To investigate the inhibition mechanism of tetramethylpyrazine combined with cisplatin on angiogenesis in Lewis lung cancer mice and to observe the mechanism of Arresten on angiogenesis in lung cancer. Methods The model of Lewis lung adenocarcinoma mouse xenograft was established in this work, and 40 mice were randomly divided into 4 groups: 0.9% sodium chloride solution group(NS group), tetramethylpyrazine group(TMP group), cisplatin group(DDP group), tetramethylpyrazine plus cisplatin group(TMP + DDP group), 10 mice in each group.Mice in NS group were given 0.2 mL of 0.9% sodium chloride solution, mice in DDP group were given 0.2 mL of 2 mg.kg-1 of cisplatin, mice in TMP group were given 0.2 mL of 100 mg.kg-1 of tetramethylpyrazine, mice in TMP+DDP group were given 2 mg.kg-1 of cisplatin and 100 mg.kg-1 of tetramethylpyrazine, each 0.1 mL .Tumor size was measured every day to calculate the tumor volume. The mice were sacrificed to stripp the subcutaneous tumor after continuous medication. The expressions of Arresten, integrin α1β1 and VEGF were determinated by immunhistochemistry and Western blotting. Results The tumor growth of NS group was the fastest and TMP+DDP group was the slowest. Compared with NS group, the expression of Arresten in the other three groups was increased( P<0.01) , and the TMP+DDP group exhibited the highest expression;at the same time, integrin α1β1 , VEGF in the other three groups was decreased(P<0.01), and the TMP+DDP group exhibited the lowest expression. The expression of integrinα1β1 and VEGF was negatively related to Arresten, and the expression of integrin α1β1 was positively correlated with VEGF. Conclusion TMP can inhibited the growth of Lewis lung carcinoma and angiogenesis. Moreover, in combination with cisplatin, TMP can also improved the effect of chemotherapy and then the survival state of mice. The mechanism of action, which TMP suppress tumor angiogenesis may be through improving Arresten and inhibiting integrin α1β1 and VEGF. And the action mechanism of Arresten may be implemented by inhibiting the expression of VEGF by incorporation with integrinα1β1 or by itself to inhibit the expression of VEGF.

RESUMO

The nanocomposites of poly-diallyldimethylammonium chloride (PDADMAC) and CdTe quantum dots (QDs) (i.e.QDs-PDADMAC nanocomposites) have been prepared based on electrostatic interaction and their fluorescence stability in aqueous solution has been investigated. MTT method(3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazoliumbromide method) was used to study their cytotoxicity and A549 lung cancer cell as a model cell was also used to evaluate their cellular imaging. It was shown that the fluorescence stability of QDs-PDADMAC nanocomposites was much better than that of bare QDs both in aqueous solution and cell.Meanwhile,QDs-PDADMAC nanocomposites display very low cytotoxicity in the low concentrations and better staining ability compared with QDs.QDs-PDADMAC nanocomposites will have great advantage on the cell analysis detection and imaging.

RESUMO

This paper studied the expert system of genotype discrimination for the STR locus D5S818 based on near-infrared spectroscopy-principal discriminant variate (PDV).Six genotypes,i.e.genotypes 10-10,10-11,11-11,11-12,11-13 and 13-13,were selected as research subjects.Based on the optimum polymerase chain reaction (PCR) conditions, about 54 measuring samples for each genotype were obtained; these samples were tested by near-infrared spectroscopy directly. With differences between homozygote genotypes and heterozygote ones, and differences of the total number of core repeat units between the six genotypes, two types of genotyping-tree structure were constructed and their respective PDV models were studied using the near-infrared spectra of the samples as recognition variables. Finally, based on the classification ability of these two genotyping-tree structures,an optimum expert system of genotype discrimination was built using the PDV models. The result demonstrated that the built expert system had good discriminbility and robustness; without any preprocessing for PCR products, the six genotypes studied could be discriminated rapidly and correctly. It provided a methodological support for establishing an expert system of genotype discrimination for all genotypes of locus D5S818 and other STR loci.

ObjectiveTo analyze the changes of imaging and the efficacy of sympathetic cervical spondylosis before and after treatment and discuss the correlation of them. Methods Seventy-one sympathetic cervical spondylosis inpatients who examined by X-ray,CT,transcranial Doppler (TCD) or color Doppler flow imaging(CDFI) were collected. After comprehensive treatment, the efficacy was evaluated according to the recovery of patients' symptoms, and the cases with imaging abnormalities were examined again. Then the correlation between the changes of imaging and the efficacy was analyzed.ResultsAfter treatment, all 71 cases, including 45 cases with excellent efficacy,18 cases with good effect and 8 cases turned effective, showed a fine rate 88.7% (63/71) and an effective rate 100.0% (71/71).The rechecked imaging results showed:in 36 lantoaxial subluxation cases by CT scanning,26 cases turned to be normal,6 cases were improved and 4 cases had finally no change; in 68 cases with abnormal X-ray imaging,67 cases were improved and 1 case had finally no change; in 38 cases with abnormal TCD results,24 cases turned to be normal,2 cases were improved,10 cases showed changes and 2 cases had finally no change;in 36 cases with abnormal cervical CDFI results,9 cases turned to be normal,10 cases were improved,15 cases showed changes and 2 cases had finally no change. The analysis of correlation between the changes of imaging and the efficacy showed:changes count by the value 0,r =0.388,t =3.500,P< 0.01 ; changes count by the value 0.5,t =0.361,t =3.211,P <0.01.ConclusionsThe efficacy of sympathetic cervicalspondylosis has positive correlation with the changes of imaging and the causes of this disease are complex. So complete and well inspection can help the syndrome differentiation treatment and confirm the efficacy.

RESUMO

Objective To investigate the effect of glycosylphosphatidylinositol specific phospholipase D (GPI-PLD) on artherosclerosis. Methods The GPI-PLD activities and mononuclear cells GPI-PLD gene mRNA expression was detected in 102 patients with coronary artery disease and 121 healthy adult. The GPI-PLD highly expressing cell model was constructed and induced by ox-LDL,the HUVEC and HUVEC transfected with pcDNA3.1(+) as blank and control group, respectively. Before and after the induction, the change of cellular function and biological features was detected. Results The peripheral blood GPI-PLD activity of patients with coronary heart disease and normal control were 31.80±4.21 and 44.32±4.50,and the IA ratio of GPI-PLD mRNA expression of mononuclear cells were 0.92±0.16 and 1.53±0.14,respectively.The activity of GPI-PLD and mRNA expression in patients with coronary artery disease were decreased up to 28.2% (t=21.31,P<0.01) and 39.9% (t=30.36,P<0.01) as compared with healthy control. The adhesion cells,ET-1,reactive oxygen, malondialdehyde (MDA) and the apoptosis rate of GPI-PLD overexpressing HUVEC were lower as compared with blank [29.59=1.40,3.51 ± 0.45,(50.63 ±4.22) ng/L,0.043±0.011,(3.32±0.44) nmol/L vs.41.39±2.15,10.57±1.12,(59.35±4.45)ng/L,0.052±0.011,(5.01±0.69) nmol/L],and as compared with control [42.68±2.45,9.92±1.03,(61.11±4.12) ng/L,0.051±0.007,(4.89±0.71) nmol/l] after inducing by ox-LDL; while the level of nitrogen noxidium (NO) was higher than blank [(29.88± 1.37)μmol/L vs.(21.76±1.02)μmol/L] and control(23.43±0.85)μmol/L groups. Conclusions The expression and activity of GPI-PLD in patients with coronary artery disease are lower than health people.Stable high expression of GPI-PLD is beneficial to vascular endothelial cell injury repairment and prevent the occurrence and development of atherosclerosis.

RESUMO

Exosomes are nanometer sized membrane vesicles, released in the extracellular milieu following the fusion of the external membrane of multivesicular body (MVB) with plasma membrane. They perform a certain function in immune regulation. Exosomes have been shown to be released by cells of hematopoietic and non-hematopoietic origin. Tumour-derived exosomes (TEX) exist in the supernatant of tumour cells, plasma and malignant effusions of tumour patients. They contain native candidate tumour associated antigen and are capable of transferring antigens to T lymphocytes, therefore efficiently promoting cytotoxic T lymphocyte (CTL) activation and producing antitumor immunity. However, recent evidence shows that tumor exosomes may induce immunologic tolerance and even activate immunosuppression which makes tumour escape from the immune surveillance of the host immune system. In addition, tumor exosomes may mediate a growth-promoting effect on tumor cells. These discrepancies are almost certainly due to differences in the phenotype of the exosomes.
